Do I have to subscribe to Wordpress.org to be able to upload the Flexibility theme? If not, how do I go about doing so and are there any tutorials available on how to use the theme?

No, you need a self-hosted WordPress installation. There are tutorials for FlexSqueeze, which you would be able to get an idea of how Flexibility 2 works, just without all the features.
